چکیده

Platelet-derived growth factor (PDGF) is a potent mitogen for cells of mesenchymal origin, and stimulates the proliferation of density-arrested Balb/c-3T3 fibroblasts (Pledger et al., 1978; Williams et al., 1982; Sturani et al., 1989; Coats et al., 1991). PDGF was first purified from human platelets and shown to consist of a heterodimer composed of two disulphide-linked polypeptide chains, designated A and B (PDGF-AB) (Antoniades et al., 1979; Heldin et al., 1979; Raines and Ross, 1982). Subsequently, two additional homodimeric isoforms of PDGF

Platelet-derived growth factor receptors on macrovascular endothelial cells mediate relaxation via nitric oxide in rat aorta.

The effects of platelet-derived growth factor (PDGF) were studied in isolated rings of rat aorta contracted submaximally to phenylephrine. The BB isoform of PDGF elicited relaxation in rings with endothelium and further contraction in rings without endothelium. Both the endothelium-dependent relaxation and endothelium-independent contraction occurred at concentrations known to induce PDGF recep...

PDGF-BB-mediated activation of p42 is independent of PDGF b-receptor tyrosine phosphorylation

Cartel, Nicholas J., Jason Liu, Jinxia Wang, and Martin Post. PDGF-BB-mediated activation of p42MAPK is independent of PDGF b-receptor tyrosine phosphorylation. Am J Physiol Lung Cell Mol Physiol 281: L786–L798, 2001.—Herein, we investigated the activity of mitogen-activated protein kinase (MAPK), a key component of downstream signaling events, which is activated subsequent to platelet-derived ...

Lack of effect of recombinant platelet-derived growth factor on human neutrophil function.

Platelet-derived growth factor (PDGF) has been reported to induce chemotaxis, degranulation, and superoxide anion generation, and to increase the expression of CD11b/CD18 in human neutrophils; hence, it has been proposed as an important regulator of neutrophil function. Most of the studies on PDGF, however, have been complicated by the use of nonrecombinant PDGF or the use of mixed leukocyte ce...

The platelet-derived growth factor receptor stimulation of p42/p44 mitogen-activated protein kinase in airway smooth muscle involves a G-protein-mediated tyrosine phosphorylation of Gab1.

Using cultured airway smooth muscle cells, we showed previously that the platelet-derived growth factor (PDGF) receptor uses the G-protein, G(i), to stimulate Grb-2-associated phosphoinositide 3-kinase (PI3K) activity. We also showed that this was an intermediate step in the activation of p42/p44 mitogen-activated protein kinase (p42/p44 MAPK) by PDGF. We now present two lines of evidence that ...

Evidence for Functional Platelet-derived Growth Factor Receptors on MG-63 Human Osteosarcoma Cells1

The specific interaction of platelet-derived growth factor (PDGF) with the human osteosarcoma cell line MG-63 was studied. Scatchard analysis of 125I-PDGFbinding to MG-63 cells indicated there were 32,000 specific PDGF-binding sites per cell with a K<jof 2.4 x 10~11M. Unlabeled PDGF blocked the specific binding of labeled PDGF to MG-63 cells at concentrations greater than 1 ng/ml. When assayed ...
